A deterministic simulation study of embryo marker-assisted selection for age at first calving in Nellore (Bos indicus) beef cattle Genet. Mol. Biol.
Rosa,Artur J.M.; Bijma,Piter; Oliveira,Henrique N.; Lôbo,Raysildo B.; van Arendonk,Johan A.M..
We used deterministic simulation of four alternative multiple ovulation and embryo manipulation (MOET) closed nucleus schemes to investigate the benefits of using marker-assisted selection (MAS) of Nellore (Bos indicus) beef cattle embryos prior to transplantation to reduce the age at first calving (AFC). We found that MAS resulted in increased genetic gain as compared to selection without AFC quantitative trait loci (AFC-QTL) information. With single-stage selection the genetic response (GR) increased as follows: GR = 0.68% when the AFC-QTL explained 0.02 of the AFC additive genetic variance (sigma2A); GR = 1.76% for AFC-QTL explaining 0.05 sigma2A; GR = 3.7% for AFC-QTL explaining 0.1 sigma2A; and GR = 55.76% for AFC-QTL explaining 0.95 sigma2A. At the...

Analytical and numerical comparisons of two methods of estimation of additive × additive interaction of QTL effects Scientia Agricola
Bocianowski,Jan.
Epistasis (additive × additive interaction) plays an important role in the genetic architecture of complex traits. This study presents analytical and numerical comparisons of two methods of estimation of additive × additive interaction of QTL effects. In the first method, we observed only the plant phenotype, while in the second method we have additional information from the molecular markers observations. In this study, two data sets were analyzed: i) 150 barley (Hordeum vulgare L.) doubled-haploid lines derived from the cross Steptoe × Morex and ii) 145 doubled-haploid lines of barley obtained from the cross Harrington × TR306. In total, 153 sets of observations were analyzed. The additive × additive interaction effect calculated on the basis of the...

Genetic linkage map of Phaseolus vulgaris and identification of QTLs responsible for resistance to Xanthomonas axonopodis pv. phaseoli Trop. Plant Pathol.
Santos,Amaury S.; Bressan-Smith,Ricardo E.; Pereira,Messias G.; Rodrigues,Rosana; Ferreira,Claudia F..
Common bean (Phaseolus vulgaris) cultivars with a high degree of resistance to Xanthomonas axonopodis pv. phaseoli (Xap) are not available in Brazil. Despite many studies, a low degree of resistance to Xap continues to exist due to its complex genetic inheritance, which is not well known. The objectives of this research were to complement a common bean genetic map based on the cross between a susceptible genotype 'HAB-52' and a resistant genotype 'BAC-6', and to map and analyze genomic regions (quantitative trait loci – QTLs) related to Xap resistance. Eleven linkage groups were determined using 143 RAPD markers, covering 1,234.5 cM of the genome. This map was used to detect QTLs associated with Xap resistance on leaves and pods. The averages of disease...

Fusarium head blight (FHB) is one of the most devastating diseases of wheat (Triticum aestivum L.), and the development of cultivars with FHB resistance is the most effective way to control the disease. Yumechikara is a Japanese hard red winter wheat cultivar that shows moderate resistance to FHB with superior bread-making quality. To identify quantitative trait loci (QTLs) for FHB resistance in Yumechikara, we evaluated doubled haploid lines derived from a cross between Yumechikara and a moderate susceptible cultivar, Kitahonami, for FHB resistance in a 5-year field trial, and we analyzed polymorphic molecular markers between the parents. Our analysis of these markers identified two FHB-resistance QTLs, one from Yumechikara and one from Kitahonami. The...

Saifert,Luciano; Sánchez-Mora,Fernando David; Assumpção,Wilson Taybar; Zanghelini,Jean Alberto; Giacometti,Renan; Novak,Eduardo Irineu; Vesco,Lírio Luiz Dal; Nodari,Rubens Onofre; Eibach,Rudolf; Welter,Leocir José.
Abstract: The objective of this work was to use a marker-assisted selection for pyramiding the resistance loci Rpv1 and Rpv3.1 in grapevine (Vitis vinifera), and to evaluate their conferred resistance against Brazilian downy mildew (Plasmopara viticola) isolates. A progeny of 23 plants, segregating for the two resistance loci, was obtained by the cross between the Gf 2000-305-122 and Gf.Ga-52-42 genotypes. The progeny was genotyped with four microsatellite markers and phenotyped for resistance to P. viticola using a bioassay with leaf discs. Six plants containing the Rpv1 and Rpv3.1 pyramided loci were identified by the molecular analysis. Plants harboring the Rpv1 + Rpv3.1, Rpv3.1, and Rpv1 loci showed 12.8, 30.0, and 33.1 sporangiophores per leaf disc,...

Soares,Taís Cristina Bastos; Good-God,Pedro Ivo Vieira; Miranda,Fábio Demolinari de; Soares,Janaína Bastos; Schuster,Ivan; Piovesan,Newton Deniz; Barros,Everaldo Gonçalves de; Moreira,Maurilio Alves.
The objectives of this study were to detect quantitative trait loci (QTL) for protein content in soybean grown in two distinct tropical environments and to build a genetic map for protein content. One hundred eighteen soybean recombinant inbred lines (RIL), obtained from a cross between cultivars BARC 8 and Garimpo, were used. The RIL were cultivated in two distinct Brazilian tropical environments: Cascavel county, in Paraná, and Viçosa county, in Minas Gerais (24º57'S, 53º27'W and 20º45'S, 42º52'W, respectively). Sixty-six SSR primer pairs and 65 RAPD primers were polymorphic and segregated at a 1:1 proportion. Thirty poorly saturated linkage groups were obtained, with 90 markers and 41 nonlinked markers. For the lines cultivated in Cascavel, three QTL...

Use of the QTL approach to the study of soybean trait relationships in two populations of recombinant inbred lines at the F7 and F8 generations Braz. J. Plant Physiol.
Vieira,Antonio José Dias; Oliveira,Dario Alves de; Soares,Taís Cristina Bastos; Schuster,Ivan; Piovesan,Newton Deniz; Martínez,Carlos Alberto; Barros,Everaldo Gonçalves de; Moreira,Maurílio Alves.
This work aimed to identify the quantitative trait loci (QTL) associated with photosynthesis and growth and productivity traits of soybean and to study possible associations between these traits by the analysis of coincidence of QTL in linkage groups (LGs). Thus, populations of recombinant inbred lines (RILs) of the F7 and F8 generations derived from the cross between the varieties BARC-8 and Garimpo were used. The traits evaluated were net assimilation rate of CO2 under saturating light (Asat), potential photosynthesis rate (Pmax), leaf area (A), specific leaf area (SLA), specific leaf nitrogen (N); root (W R), nodule (W N), stem (W ST), leaf (W L), pod (W P) and plant dry mass (W T); nodule (nN), seed (n s), and pod number (nP); seed fresh mass per plant...

What role for genomics in fisheries management and aquaculture? ArchiMer
Wenne, Roman; Boudry, Pierre; Hemmer-hansen, Jakob; Lubieniecki, Krzysztof P.; Was, Anna; Kause, Antti.
The development and application of genomics has been facilitated in a number of fields by the availability of new methodologies and tools, such as high throughput DNA sequencing and complementary DNA (cDNA) microarrays. Genomic tools are already used in research on commercially important fish and shellfish species. Thousands of expressed sequence tags (EST) are now available for some of these species, and the sequencing of complete genomes of tilapia, cod, salmonids, flatfishes, sea bass and Pacific oyster has been proposed. Microarray technology through simultaneous analysis of the expression of thousands of genes allows the identification of candidate genes involved in the function of multiple physiological, morphological and behavioural traits of...
